Pipette solution was 100 mM KMeSO₃, 1 mM MgCl₂, 1mM EGTA, 25 mM (NH₄)₂SO₄, 10 BES and 1 mg/mL amphotericin at pH 7. The bath solution was 100 mM TMAMeSO₃, 1 mM EGTA, 2 mM MgCl₂, 1.5 CaCl₂, 25 mM (NH₄)₂SO₄, and 10 mM BES pH 7. At the times indicated by the arrows, the cell was stimulated with 60 nM PMA and I_e was inhibited by 12 μM DPI.
