(A) V5-tagged IKKε and FLAG-tagged wt or the indicated C-terminal truncated mutants of IKKε were co-transfected into 293T cells. Cells were lysed 24 h post-transfection. Lysates were immunoprecipitated with FLAG-agarose and V5-tagged IKKε were detected with an anti-V5 antibody (top panel). V5 and FLAG-tagged IKKε in whole cell lysate (WCE) were also detected as indicated on the left. (B) V5-tagged IKKε and FLAG-tagged wt or IKKε mutants with a single amino acid substitution, as indicated at the top, were co-transfected into 293T cells. Immunoprecipitation was performed as in (A).
