Figure 2. TLR4 mediates the phosphorylation of IRF3 in response to HSV-2.

HCE cells were transfected with scrambled shRNA control or iTLR4. Forty-eight hours after transfection, cells were infected with HSV-2 at 3 MOI or mock-infected. Cell lysates were collected at 0 h, 4 h, 6 h, 16 h post-infection and analyzed by Western blotting with antibodies against p-IRF3, total IRF3 and Actin. Knockdown of TLR4 decreased the phosphorylation of IRF3 by HSV2 infection. The experiment was repeated three times, the representative images from one experiment are shown.
