(A) Diagrams illustrate genomic regions of the indicated strains and required steps for the construction of tpi mutants: (1) integration of a plasmid containing a second copy of tpi driven by the predictive native promoter into the L5 mycobacteriophage attachment (attL5) site; (2) deletion of the native tpi copy by homologous recombination generating the Δtpi::attL5-tpi strain. (B) Replacement of the tpi attL5 site copy generated the tpi-TetON mutant, in which tpi transcription is induced by anhydrotetracycline (ATC) delivered on a filter in the center of the plate.
