Apoptosis induced by CDDP/F-ara-A combination treatment is mediated by Noxa. (a) Protein expression levels of Noxa and actin in untransfected cells and in cells nucleofected with Noxa or control (scr) siRNA, untreated (left panels) or treated with 10 μm CDDP and 10 μm F-ara-A for 48 h (right panels), were analyzed by western blot for control of knockdown of Noxa. Apoptosis was assessed by MitoTracker staining (patient no. 1). (b) Protein expression levels in four patients (patient nos. 1–3 and 5), treated as described in (a) were quantified using Licor Odyssey software and the Noxa/Mcl-1 ratio was calculated.
