BLUP103 (Pspac-plsC) cells grown ON at 37°C in MM supplemented with 0.2 mmol/L IPTG were washed and used to inoculate fresh media, in the presence (PlsC+) or absence (PlsC) of 1 mmol/L IPTG. At DO600 of 0.35 cultures were split and incubated at 25 or 37°C for 6 h. When growth stopped because of PlsC depletion, total lipids were extracted, transesterified to yield FA methylesters and subjected to GC-MS analysis.
