L655,240 inhibits the activity of BACE1 but not α-secretase or γ-secretase. HEK293-APPswe cells were incubated with L655,240 for 24 h before harvesting. (A) L655,240 inhibited BACE1 activity in HEK293-APPswe cells in a concentration-dependent manner. (B) L655,240 had no effect on either γ-secretase activity or ADAM10 expression. Cell lysates were subjected to Notch and ADAM10 analysis by Western blot (upper panel); the quantitative results are also shown (lower panel).
