Triptolide inhibits the expression of NF-κB and the accumulation of p65 in nucleus. (A) Cells were treated with triptolide at the indicated concentrations for 24 h. The nuclear and cytoplasm fractions of cells were harvested, and then were analyzed by Western blotting using anti-nuclear factor κB (NF-κB) p65 and phos-p65 antibodies. The blots were blotted with actin or lamin B antibody to control for sample loading. Results shown are representative of three separate experiments. (B) Cells treated with 40 nmol/L of triptolide were fixed, permeabilized, and then subjected to immunofluorescent double staining with antibody against p65 subunit of NF-κB by Texas Red. Nuclei were counterstained with DAPI.
