Dau inhibited p70S6K and 4E-BP1 phosphorylation and rapamycin, a specific inhibitor of mTOR, suppressed IGF-I-induced HIF-1α protein accumulation. (A) Serum-starved MCF-7 cells were pretreated for 1 h with different concentrations of Dau, followed by incubation with IGF-I (40 ng/mL) for 6 h. Phosphorylated p70S6K and 4E-BP1 levels were determined by Western blotting. (B, C) Serum-starved MCF-7 cells were pretreated with various concentrations of rapamycin for 1 h, followed by incubation with IGF-I (40 ng/mL) for 6 h. HIF-1α protein levels were analyzed by Western blotting (B), and VEGF production in the conditioned medium was determined by ELISA (C).
