Rescue of fusion-promoting activity of attachment and fusion-deficient mutants by coexpression with an hPIV3-NDV HN chimera. The figure shows the fusion-promoting activity exhibited by NDV-AV mutants I175E, D198R, K236R, Y526L, and E547Q. The mutated HN proteins carry an innocuous mutation (E347G) that renders them unrecognizable by an antibody to antigenic site 14. Cells coexpressing the mutated HN protein, the rescuing chimera, and the NDV F protein are pretreated for 30 min at room temperature with 200 μg of site 14 antibody in 100 μl of medium just before mixing with the target cells in the reporter gene content-mixing assay. Data are expressed as percentages of wt HN carrying the site 14 mutation and treated in the same way.
