Budding of viruses by thin-section electron microscopy. MDCK cells grown on polycarbonate filters were infected with different viruses at an MOI of 3.0. At 12 h p.i., infected cell monolayers were cross-linked in 2% glutaraldehyde and postfixed with 1% OsO4. Filters were embedded in Epon, and 60-nm-thick sections were stained with uranyl acetate and lead citrate and examined with a transmission electron microscope.
