CMS5 cells expressing RAP or control protein were transfected with gp96-EGFP. Robust fluorescence of gp96-EGFP was analyzed by flow cytometry (A) and confocal microscopy (B). The foot pad of BALB/c (C), or CD91−/− (D), mice were implanted with 5×106 of either cell type and the draining or contralateral non-draining lymph nodes were harvested two days later. Cells were stained for the hematopoietic marker CD45 and analyzed by flow cytometry for EGFP. The EGFP signal was normalized against the starting fluorescence in each cell type and the background fluorescence of non-draining lymph nodes. *** P < 0.001, n.s. not significant. Experiments were independently performed twice with 8 mice per group (A), or 3 mice per group (B). Error bars indicated s.e.m.
