Thioflavin (ThT) fluorescence assay of peptide chaperones. Peptides were incubated at 37 or 42 °C for 72 h as described in Materials and Methods. ThT (final concentration of 10 μM) was added following incubation with or without shaking, and the fluorescence spectra were recorded in a Jasco-750 spectrometer after excitation of the samples at 350 nm. (A) ThT fluorescence of mini-αA-chaperone, suggesting the presence of fibril-like structures. (B) Absence of ThT fluorescence with the CP1 peptide, indicating the absence of fibril-like structures.
