Augmented IFNγ production and reduced IL-17 production in T cells from WW donors. A, Purified CD4+ T cells from donors with indicated genotypes were stimulated with T cell expander beads (bead:cell ratio 1:1) for 20 hours, followed by measurement of various cytokines in the supernatants (RR; filled circles, RW; filled triangles, WW; open circles); *P < 0.05, **P < 0.01, ***P < 0.001, determined by t-test or Mann-Whitney Rank Sum Test. B, Experiment as in A, but with different bead:cell ratios. Data are presented as average ± SEM (three donors of each genotype, RR; filled circles, WW; open circles). C, Purified T cells were stimulated for five hours with PMA/ionomycin, the last four hours in presence of Brefeldin A. Later, levels of intracellular cytokines in various CD4+ subsets were assessed by FACS analysis. Data are presented as average ± SEM (RR; black, WW; white). One donor pair (consisting of one RR donor and one WW donor) was analyzed in each experiment and a total of four experiments were conducted. Due to variation in staining with cytokine-specific antibodies from one day to another, paired t-test was used for statistical analysis; *P < 0.05, **P < 0.01, determined by t-test.
