IL-15 regulates caspase-3 activity independently of caspase-8 and caspase-9. (a) Day 5 IL-2- and IL-15-cultured T cells were lysed in buffer containing biotin-VAD (bVAD) to selectively label active caspases. Then, 450 μg protein lysate was selectively precipitated with streptavidin-sepharose beads. Precipitates were resolved by SDS-PAGE and immunoblotted for caspase-8, caspase-9, and caspase-3, and compared with whole-cell lysates (WCLs; 30 μg) from the same cells.
