Effects of Nrf2 knockdown on autophagy level in A2780cp cells. A. mRNA was extracted from A2780cp cells after transiently transfecting them with control siRNA or Nrf2 siRNA (100 nM) for 48 h. After the mRNA was converted into cDNA, the cDNAs were subjected to real-time quantitative PCR for analysis of relative mRNA levels of Atg3, Atg5, beclin 1, Atg12, p62 and actin. Data was normalized to actin level. B. Cell lysates from A2780cp cells transfected with control siRNA or Nrf2 siRNA (100 nM) for 48 h were collected for western blot analysis for Atg3, Atg5, beclin 1, Atg12 and p62. C. Autophagosomes were detected by transmission electron microscopy after the cells were transfected with control siRNA or Nrf2 siRNA (100 nM) for 48 h (Magnification was 10000×). Scale bars are 1 μm.
