(A) No LA was detected in any region of left ventricle in the saline-treated animals on day 3 or day 14 (n=4). In the Ad5Luc group, the ischemic zone exhibited robust LA, which was significantly greater than that in the border zone adjacent to the AAR (***p<0.001, n=4). At day 14, the ischemic zone still demonstrated the strongest LA compared to nonischemic zone or border zone (n=8). However, the LA at day 14 was significantly reduced in infarct and border zones compared to that on day 3 (***p<0.001, n=8). Values are presented as mean±SEM. (B) PCR detection of Ad5Luc DNA in myocardial tissues. The tissues were obtained from three animals (animals 4, 5, and 6). The samples were obtained 3 days after intracoronary delivery of Ad5Luc. Tissues that were obtained from the AAR showed strong adenoviral transduction in all three animals. The PCR product from the border zone (BZ) showed a weak signal. Nonischemic zone (NIZ) displayed no signal (animals 4 and 6) or very weak expression (animal 5).
