Michaelis-Menten plots for purified CAT: (A and D) untagged; (B and E) fused to a C-terminal PDZ2 domain peptide ligand; and (C and F) fused to an N-terminal PDZ2 domain. Transferase activity of purified CAT was assayed with (A-C) 500 μM Acetyl-CoA and 0-100 μM Chloramphenicol, or with (D-F) 100 μM Chloramphenicol and 0-500 μM Acetyl-CoA for 10 minutes at 37 °C. Michaelis-Menten constants (Table 5), were calculated as described in Methods.
