Figure 7. Knockdown of Beclin1 or ATG5 protects cells from pazopanib and valproate toxicity. HT1080 cells were transfected with scrambled control siRNA (siSCR) or siRNA molecules to knock down expression of Beclin1 (siB1) or ATG5 (siA5). Thirty-six hours after transfection cells were treated with vehicle (DMSO), pazopanib (P, 5.0 μM), and/or valproate (V, 500 μM). Cells were isolated 24 h after exposure and viability determined by trypan blue exclusion (n = 3, ± SEM). #P < less than corresponding value in siSCR cells.
