HERC2 regulates p53 activity in a proteasome- and MDM2-independent manner. Lysates from transfected U2OS cells with NT or HERC2 siRNAs were analyzed by immunoblotting with antibodies against the indicated proteins. Before lysis, cells were treated with the proteasome inhibitor MG132 (A and C) or with the MDM2 inhibitor Nutlin 3a (B) for 6 and 16 h, respectively. Data are representative of at least three independent experiments.
