Overexpression of mortalin protects K562 cells from complement-dependent cytotoxicity. A, K562 cells transfected for 48 h with a mortalin-EGFP or EGFP plasmid were analyzed under a confocal microscope. A representative cell of each is shown in fluorescence and bright field. Transfected or nontransfected cells were treated with anti-K562 antibodies and NHS for 1 h at 37 °C. B, next, the cells were washed, mixed with propidium iodide, and analyzed by flow cytometry. Percentage of cell lysis was determined based on the percentage of cells labeled with propidium iodide. For correction of specific cell lysis, percentage lysis after treatment with HIS was subtracted from that of cells treated with NHS. *, p < 0.01; **, p < 0.001. Error bars, S.D.
