Simultaneous inhibition of cancer cell invasion (left) and TNFα gene delivery (right) by VGD polyplexes in U2OS cells. U2OS cells were treated with VGD/DNA polyplexes prepared with TNFα plasmid DNA. The VGD6 and VGD10 polyplexes were prepared at N/P 8 and VGD8 polyplexes were prepared at N/P 4. The cells were allowed to invade through a layer of Matrigel upon stimulation with SDF-1 for 16 h. AMD3100 (300 nM) was used as a positive control. TNFα gene delivery efficiency of VGD was determined from cell viability measured by MTS assay; normalized to cell viability observed with polyplexes formed with luciferase DNA.
