M2′ and M2″ remain bioactive to human colon cancer cells but less toxic to human normal colon cells than 8S and 10S and M2 induces apoptosis on HCT-116 and HT-29 human colon cancer cells. Growth inhibitory effects of 8S, M2′, 10S, and M2″ against human normal colon cells CCD-18Co (A) and human colon cancer cells HCT-116 (B) and HT-29 (C) treated with 8S, M2′, 10S, or M2″ for 24 h at different doses (n = 6) (measured by MTT assay). (D) Effect of M2 on apoptosis in human colon cancer cells HCT-116 and HT-29 treated with M2 (0, 10, 20, and 40 μM) for 24 h measured by PI staining; data represent mean ± SD. (E) Western blot analysis of HCT-116 and HT-29 cell extracts treated with DMSO, 10, 20, and 40 μM of M2 for 24 h. Fold induction for each marker compared with DMSO is indicated under the corresponding line.
