Absolute quantitation of an albumin ¹⁴C-iodoacetamide modification competitive binding experiment. Treated and untreated BSA samples were exposed to 1 pCi ¹⁴C-iodoacetamide and described as follows: ¹⁴C-iodoacetamide-modified native BSA (¹⁴C-IAC-native):native BSA with 1 free Cys residue; ¹⁴C-IAC-native + GSH:native BSA and 5 mM GSH; ¹⁴C-iodoacetamide-modified reduced BSA (¹⁴C-IAC-reduced):disulfide bonds reduced; ¹⁴C-IAC-reduced + GSH:disulfide bonds reduced and 5 mM GSH. Results were background subtracted from the control, which showed no excess ¹⁴C above the endogenous level expected for a contemporary biological sample. Error bars represent the standard error of the mean of three replicate samples.
