ELMOD1–3 GAP activities were determined using ARL2·[γ-³²P]GTP as substrate with the charcoal GAP assay. A fixed amount of substrate was used in these assays and the amount of GAP was varied to generate the dose-response curves shown. Duplicate time points were taken and averaged, with error bars representing mean ± S.E. Note the differences in the y axes with GST-ELMOD1 (left), GST-ELMOD2 (center), and GST-ELMOD3 (right) due to the large differences in specific activities of the three GAPs for this one GTPase.
