(A) Time courses of pAKT, F16P, PEPCK, glycogen and GLCex in response to a step stimulation (red), a ramp stimulation (blue) and 0 nM (black) of insulin in FAO cells. The mean values and SEMs of three independent experiments are shown. The step and ramp stimulations of insulin are shown at the bottom. The final concentrations of step and ramp stimulation of insulin were set at 0.1 nM, which mimics the basal secretion of insulin in vivo (Polonsky et al, 1988b; Basu et al, 1996). Note that the scale of absolute concentration of GLCex was normalised because of the variation between experiments.
