YukE Western blot analysis of the control cell lysates (C1) and medium supernatant (S1) samples, BMOE-treated cells (C2), cells (C3) recovered in the absence of crosslinker, and newly secreted proteins (S3). The dimeric YukE species, crosslinked while inside the cells, is present in the final supernatant (S3) sample even after adding the excess of reducing agent DTT.
