Dex completely reversed the immunosuppressive effects of MSCs in vitro. (a) Splenocytes from C57BL/6 mice were co-cultured with MSCs from WT C57BL/6 mice (1 : 20 ratio of MSC to splenocytes) in the presence of anti-CD3, with or without Dex, and proliferation was assayed by 3H-Tdr incorporation. (b) Total nitrates in the supernatant of the cultures from (a) were assayed by modified Griess reagent. (c and d) Proliferation of human PBMCs co-cultured with human MSCs (1 : 10 ratio of MSCs to PBMCs) in the presence of anti-CD3 and anti-CD28, with or without Dex or methylprednisolone, respectively.
