Overview of multiplexed direct genomic selection (MDiGS) methodology. Three BACs were selected for use as baits and biotinylated for targeted capture of 554 kb of coding and non-coding sequence surrounding PITX1, 5′ HOXC genes and TBX4. Genomic input DNA (150 ng) was individually indexed and pooled in batches of 96. Multiplexed DNA pools were hybridized with biotinylated BAC baits and enriched target regions are sequenced on MiSeq Personal Sequencer.
