The RNA incorporating 5-FU metabolite induces phosphorylation of eIF2α. (A) HeLa cells were treated with increasing concentrations of the 5-FU metabolites FUrd or FdUrd for 72 h. Cell lysates were prepared and intracellular levels of eIF2α and P-eIF2α were analyzed. Equal loading was verified by Coomassie gel staining. (B) HeLa cells were treated with the indicated concentrations of FUrd or FdUrd for 72 h and processed for immunostaining of phosphorylated eIF2α (red) and the SG marker protein TIAR (green).
