SMAD2 and SMAD3 mRNA 3′UTR contains a binding site for miR predicted by TargetScan (A–B). Increased expression of miR-146a in diaphyseal cell populations was correlated with reduced expression in SMAD3 mRNA levels but not SMAD2 mRNA levels (D–E). Increased expression of miR-146a in diaphyseal cells was coupled with an increase in expression of RUNX2 and ALP (F–G). Reduced expression of SOX9 was observed when miR-146a was highly expressed. Relative expression was normalized to MAMM-U6 (C) and β-Actin (D–H).
