Three cryoLM and cryoEM data sets were acquired on a vitrified cellular sample infected with Ad5-488 viruses. TetraSpeck microspheres of 200 nm diameter were used to measure the alignment precision σA between green and blue images. FluoSpheres of 100 nm were used for the fiducial based alignment. The correlation inaccuracy σC was calculated and the inaccuracy of locating the event of interest σE was measured by applying a 2D Gaussian fit. The overall total correlation inaccuracy for each event of interest was calculated as σT=√((σA)2+(σC)2+(σE)2). The number of TetraSpeck* microspheres and FluoSpheres** used for each experiments are indicated in brackets.
