Validation of GFP and mCherry signals in IRF sections by spectral profiling. (A, B) Spectral profile of GFP (A) and mCherry (B) in fixed and mounted cells, and in each of the three resins, showing normalised intensity across the lambda stack. No significant shift in emission profiles was noted aside from changes in background noise characteristics. (C, D) The bleaching rates of resin embedded GFP (C) and mCherry (D) were compared to fixed and mounted cells; the fixed cells showed much higher rates of fluorophore bleaching over time.
