(A) GUS enzymatic activity of pSiM24 and pSiM24-GUS A. tumefaciens-infiltrated N. benthamiana leaves was measured, and one unit of GUS activity was defined as the amount of enzyme that liberated 1 pmol 4-methylumbelliferone mg−1 protein min−1. The data represent means ± S.D. of four biological replicates for each construct (n = 4). The values significantly differ between control (pSiM24) and pSiM24-GUS agro-infiltrated leaf samples at P<0.01 based on Student's t-test. ILw: Whole Infiltrated Leaf; ILp: Partial Infiltrated Leaf. (B) The pSiM24 and pSiM24-GUS A. tumefaciens-infiltrated N. benthamiana leaves were histochemically stained for GUS enzyme. The pSiM24-GUS agro-infiltrated leaves showed stronger GUS expression, as detected by histochemical staining. Control: N. benthamiana leaf infiltrated with A. tumefaciens harboring pSiM24 construct; ILw: Whole Infiltrated Leaf; ILp: Partial Infiltrated Leaf.
