A, Diagram of gating strategy to exclude B cell precursors and class-switched memory B cells prior to B cell subset identification using co-expression of CD21 and CD24. Identified are transitional 1 (T1), transitional 2 (T2), naive follicular mature (FM), and marginal zone plus marginal zone precursor (MZ) subsets. B-E, Mononuclear cells from indicated BALB/c mouse and human tissues (spleen, peripheral blood [PB], and bone marrow [BM]) were stained for flow cytometry to detect CD19, IgM, CD21, and CD24. CD19+IgM+ cells falling in lymphocyte light scatter were gated and B cell subsets as diagramed in A were gated.
