Mesoscale structure formation and microtissue stiffness were not significantly altered by inhibiting collagen cross-linking or cellular contractility. Inhibition of microtissues’ contraction force (B) using low dosage blebbistatin (Blebbi) (2µM and 5µM) or inhibition of collagen cross-linking using low dosage BAPN (50 µg/mL and 100 µg/mL) did not significantly affect the microtissues’ cross-section area, α-SMA expression, collagen structure (A) or the elastic modulus (C). Under high BAPN concentration (1mg/mL), the collagen structure is less continuous (A) and the microtissue elastic modulus reduced by 25% (C). The collagen and α-SMA images are 3D views reconstructed from confocal stacks. Sample size for each condition: N > 9. ** p < 0.003; * p > 0.16 as compared to untreated condition by unpaired t-test.
