T84 or HT29 IECs were pretreated with (A) 5μg/ml Tunicamycin or (B) 4mM Benzyl-GalNAc before whole cell lysis and western blotting with NS19-9. (C) Protein lysates isolated from T84 IECs were treated with neuraminidase before immunoblotting with NS19-9. (D) Representative immunoblot demonstrating that transfection of HT29 IECs with CD44 gene silencing plasmids (shRNA1-3) decreased the expression of sLea detected by NS19-9 as compared to scrambled control.
