This study shows that Tcf3 induction in response to wounding is dependent on Stat3 and that Lcn2 is Tcf3’s downstream effector that promotes cell migration and wound repair. While Tcf3 is sufficient for the induction of Lcn2, it is not absolutely required, mostly likely because Tcf4 can compensate for Tcf3, as it does in other skin related phenotypes, or because additional factors can induce Lcn2. While Lcn2 is necessary and sufficient for proper wound repair, it does not fully explain how the loss of Stat3 impacts wound repair, because Tcf4 induction is not controlled by Stat3 and Lcn2 application on Stat3 cKO wounds does not fully rescue the repair defect.
