IL4-DCs and IL15-DCs were incubated in the absence and presence of 10 μg/ml Pam3Csk4 (TLR2/1), 25 μg/ml poly (I:C) (TLR3), or 1 μg/ml LPS (TLR4) for 20 h. Cells were harvested, and surface expression of IL-15 (sIL-15), CD83, HLA-DQ, HLA-DR, and HLA-E was determined by flow cytometry. Filled histograms depict labeling of IL15-DCs for indicated antigens (dark gray) or appropriate isotype controls (light gray). Open histograms depict labeling of IL4-DCs for indicated antigens (solid line) or appropriate isotype controls (dashed line).
