E^rns ΔC-term is not affected by heparin treatment at 37°C and is unable to bind to the cell surface at 4°C. E^rns was either preincubated in the presence of 0.1 mg/ml heparin for 15 min (heparin block) or left untreated before incubation on BT cells at 37°C (A) or 4°C (B) for 30 min. Unattached protein was removed by washes with medium (medium wash) or with medium containing 0.1 mg/ml heparin (heparin wash). Incubation of the cells in the presence of poly(I·C) at 37°C for 20 h was followed by extraction of the cytosol to evaluate Mx and β-actin expression by Western blotting. Typical results out of three independent experiments are shown.
