The expression of MAGI-1 induces apoptosis in HeLa cells but not in HT1080 cells. (A) HeLa cells were seeded on glass coverslips and transfected either with wild-type or mutant K499E MAGI-1 expression plasmids. After 48 h, the cells were fixed and incubated with the anti-FLAG antibody and counterstained with rhodamine-conjugated secondary antibodies. After incubation with the indicated antibodies, cells were processed for the identification of TUNEL-positive cells using the in situ cell death detection kit according to the manufacturer's instructions. Confocal images were taken at 480- and 510-nm wavelengths. (B) HT1080 cells were seeded on glass coverslips, and the immunodetection of MAGI-1 and the TUNEL assay were performed as described for panel A.
