Expression of mRNA in hCSSCs and hCFs during culture on patterned silk films. Expression levels of keratocyte genes were determined by qPCR as described in Methods. Bars show the relative abundance of gene-specific mRNA in comparison to 18S RNA. Black bars (▬), 0 wks (i.e., control cells at time of plating); red bars (▬), 3 weeks culture; green bars (▬), 6 wks; yellow bars (▬), 9 wks culture. “No RGD” and “RGD” refer to hCSSCs seeded on patterned silk films without and with RGD surface modification. hCFs refers to hCFs grown on RGD-modified patterned silk film. Error bars show SD of three independent samples. Expression of each gene significantly increased in hCSSC the cultured cells (colored bars) compared to the starting cells (black bars) (p < 0.05). Expression of KERA, CHST6, and ALDH in hCSSC+RGD was significantly greater than expression in hCF at all time points, whereas expression of alpha-SMA and EDA-FN was significantly less (p< 0.05).
