Peptide A-specific CTL clone established from anti-EML4-ALK CTL. (A) Peptide A-specific CTL clones established using CD107a single cell sorting. Peptide A-specific CTLs (1×105) were incubated with peptide-pulsed T2 cells (5×104) with CD107a-specific antibodies for 3.5 h at 37°C. CD8+CD107a+ cells were sorted using a FACSAria II cell sorter. Square, CD8+CD107a+ cells that are peptide A-specific CTL clones. (B) Recognition of peptide-pulsed T2 cells by peptide A-specific CTL clones. A peptide A-specific CTL clone (1×104 cells) was incubated with stimulator cells that had been pulsed with 1 μM peptide A or HIV-gag peptide. IFN-γ-producing CTLs were detected by IFN-γ ELISPOT assay. CTLs, cytotoxic T cells.
