Cells transfected with sh-BANCR or BANCR plasmid were treated with U0126 or SP600125 respectively. Western blot was performed to detect total and phosphorylation changes of ERK1/2 and JNK. U0126 or SP600125 induced significant inactivation of ERK1/2 (A) and JNK (C). Cell proliferation decreased significantly after 72 h (B, D). Note the notable effects induced by combined treatment with shRNA and inhibitors. BANCR activated ERK1/2 (E) and JNK (G) pathways. Inactivation of ERK1/2 and JNK were rescued by overexpression of BANCR. Inhibitory proliferation induced by ERK1/2 and JNK inactivation was ameliorated by BANCR (F, H).
