ATP hydrolysis and U4/U6 unwinding were monitored as described.42 For comparison of apparent unwinding efficiencies, ATP hydrolysis and U4/U6 unwinding were measured under identical conditions (200 nM yBrr2, 50 nM U4/U6 duplex, without or with 0.75 µM of the indicated yPrp8Jab1 variants, 30 °C).
