Combined effects of cyto D treatment and gelsolin gene deletion on infarct size after MCA occlusion. Gsn+/+ and gsn–/– mice were treated with cyto D (1 μg intracerebroventricularly 10 min before ischemia) or vehicle (V), respectively, and then subjected to 2 h of left filamentous MCA occlusion and 22 h of reperfusion. Cerebral infarct volume was determined quantitatively as described (22, 24). Results were confirmed with the indirect method to determine infarct size as follows: 72.1 ± 4.9 mm3 (V/gsn+/+) vs. 44.7 ± 8.2 mm3 (cyto D/gsn+/+) vs. 100.4 ± 13.1 mm3 (V/gsn–/–) vs. 45.4 ± 11 mm3 (cyto D/gsn–/–).
