Canonical hedgehog (Hh) signaling in cardiomyocytes. Neonatal rat ventricular myocytes (NRVMs) were cultured as described in materials and methods for 24 h and then switched to low serum medium supplemented with 2.5 mg/l sonic hedgehog (Shh) or 5 μmol/l purmorphamine (PUR) alone or in the presence of 5 μmol/l KAAD-cyclopamine (KAAD-CP). After 24 h, total RNA was isolated and processed for quantitative RT-PCR of Gli1 and the housekeeping gene ribosomal protein S15 (S15).
