DNA oligos required to assemble the MedLoop motif (as in Fig. 1) and P4–P6 domain of the Tetrahymena ribozyme (as in Fig. 2) as well as three single nucleotide mutants of each constructs. The fluorescent primer used in the protocol for reverse transcription is shown in the last row. Sequence color codes are the same as in Figs. 1 and 3; mutations are colored in orange. All sequences are shown from 5′ to 3′. Abbreviations for modifi cation are those used by Integrated DNA Technologies (IDT): /5–6FAM/, 5′-6-fluorescein. For the P4–P6 domain, to avoid confusion, mutant positions are given with no offset (i.e., this numbering does not correspond with the nucleotide numbering of the full group I intron of the Tetrahymena ribozyme)
