In vivo conversion of dehydroepiandrosterone and pregnenolone with B. megaterium ATCC13368 and B. megaterium ATCC13368 Δcyp106a2 whole cells. Conversions were performed using 500 μl samples of fresh 16 h cultures incubated with 200 μM of the respective steroid for 60 min at 30°C and vigorous shaking in 1.5 ml reaction tubes. Reaction was quenched and extracted with 500 μl ethyl acetate. DHEA was hydroxylated into 7β-OH-DHEA and 7α-OH-DHEA (a), and PREG was converted into 7β-OH-PREG and an unknown product P2 (b). The cyp106A2-knockout strain converted neither DHEA nor PREG (a,b).
