TrfA protein binds one specific strand of ssDNA in the DUEoriV of the origin. The interaction of TrfA and DNA was analyzed with an EMSA (A) and SPR (B) as described in Materials and Methods. (A) Increasing amounts of TrfA protein (5, 10, 15, 20 pmol) were incubated with 1 pmol fluorescently labeled dsDNA containing oriV, ssDNA of DUEoriV top strand or ssDNA of DUEoriV bottom strand as noted.
